S.P.B instigated singing as a hobby during his childhood days. Music was always a part of his interest since he was a child, who made him learn notations, and to play instruments like harmonium and flute on his own listening to his father. His father wanted him to go along with the engineers who made him join JNTU after which he discontinued his course due to typhoid and joined the Associate Member of the institution of engineers, Chennai. In the due course, he won many awards and recognitions at many singing competitions.
His first chance to enter the film industry was when he performed for amateur singers' competition and won the first prize, wherein he was offered a chance by the music director, S.P.Kodandapani. S.P.B made his first debut as a playback singer on December 15, 1966 with Sri Sri Sri Maryada Ramnna, a film scored by S.P.Kodandapani. He has sung in many languages including Telugu, Tamil, Kannada, Hindi, Malayalam and Marathi.
S.P.Balasubrahmanyam's first break as a playback singer was when he made his debut in the Hindi movie Ek Duje Ke Liye, for which he received the National Film Award for the best male playback singer.
